Basic scientific achievements

The dissertaion deals with diagnostics and identification of viruses at the molecular level in tomato plants (Solanum lycopersicum L.) widespread in Azerbaijan, and the results of the study of structural and functional alterations in plants induced by viral infections. During numerous monitorings conducted in various regions of Azerbaijan, characteristic symptoms of viral diseases were detected. Based on the results of immunochromatographic test and immunoenzymatic analysis (IEA), RNA viruses, such as Tobamovirus, Cucumovirus, Tospovirus, Crinivirus, Curtovirus, Nepovirus, Potyvirus, Potexvirus, DNA viruses Geminivirus, and mixed infections of Tobamovirus were detected in tomato plants. As a result of RT-PCR analysis carried out by using universal and specific primers, Tobacco mosaic virus (TMV), Tomato mosaic virus (ToMV), Pepper mild mottle virus (PMMoV) were identified at molecular level in samples taken from Absheron peninula and Salyan region, Tobacco etech virus (TEV) in the samples taken from Guba and Khachmaz regions and Tomato spotted wilt virus (TSWV) in samples collected in the Masalli region. As a result of PCR and sequence analysis of amplification products conducted with TY1(+) and TY2(-) primers, molecular diagnostics of Germiniviruses was performed at the species level for the first time in Azerbaijan. As a result, tomato samples collected in the Absheron peninsula were found infected with Tomato yellow leaf curl virus (TYLCV) and genomic nucleotide sequences of the isolate were placed in GenBank with accession number KY412552. Moreover, the changes occurring in the antioxidant defense system and photosynthetic apparatus were studied in virus-infected tomato plants. The studies revealed that viral infections lead to significant differences in activities and electrophoretic spectra of antioxidant enzymes (APO, BPO, GPO). A new isoform of ascorbate peroxidase enzyme and two new isoforms of benzidine-peroxidase and guaiacol peroxidase were induced in leaves during pathogenesis. In leaves of 24 tomato plants sharp increases in the amounts of glycine betaine and H2O2 which is considered to be a stress indicator was accompanied by the inhibition of the synthesis of photosynthetic pigments under the effect of Tobamovirus infections and their mixed forms. Photochemical efficiency of PSII and synthesis of its 47; 33; 29-24 and 17-15 kDa proteins decreased significantly, which is attributed to the changes in the molecular structure of PSI and in the electron transport systems of PSI and PSII. Based on the phytopathological and statistical analyses, it was concluded that the rate of spread and severity of viral diseases in tomato plants in the Salyan region was more acute compared to other vegetable growing regions of Azerbaijan
