Basic scientific achievements

1. In 1980s, some structural an evolutionary peculiarities of repeated DNA sequences of human, animal and plant species were revealed by the comparative computer analysis. It was suggested that these repeats covering a significant portion of eukaryot genomes may participate in regulation of transcription of the neighbour genes/genome regions; later, this suggestion was confirmed by multiple experimental findings.

2. To search for transcription start sites and regulatory elements, pre-mRNA polyadenylation sites in human, animal and higher plant genomes, various computer analysis tools of high accuracy (PromH, PromHP, TSSP-TCM, nsite, nsiteM, nsiteH, POLYAR), as well as PlantProm database of plant RNA polymerase II promoters have been developed.

3. In Arabidopsis and rice, it was revealed that the non-stop transcription of neighbour nuclear genes located in the “Tail-to-Head manner and the following alternative splicing and translation of the corresponding transcripts may produce chimeric polypeptides.

4. Distribution spectrum and scale of DNA sequences of the plastid and mitochondrion origin in the nuclear genomes of various higher plants (Arabidopsis, rice, wine grape, blackcottonwood, corn, sorghum) was clarified, as well as some facts indicating the expression of some intact nuclear copies of plastid and mitochondrial genes were revealed.

5. Using about 84000 promoter sequences with the experimentally determined transcription start site from 6 species including humans, (a) a new classification of RNA polymerase II promoters consisting of 3 classes (CpG, non-CpG / TATA and non-CpG / non-TATA) was given and their numerous characteristics were identified, and (b) based on this knowledge, a new high-precision promoter search method, TSShm, was developed by applying the statistical analysis approach of neural networks.

6. Applying TSShm and other computer programs, it was found that for 650 (approximately 93%) of the 702 known genes associated with cancer, there is at least 1 potential bidirectional promoter.
